I think Frieren is the hero because she was taught and is learning from Himmel and Fern is the mage because she was taught by Frieren and Stark is the warrior because of Eisen which would leave Sein as the Priest because he is one and his brother who taught him has connections to Hieter

She was referring to how Sein was stubbornly refusing to go on an adventure due to his own personal doubts, just like how Frieren refused to go on the adventure when Himmel first asked her. It's why she was determined to convince him, since she knows going on the adventure is the right thing for him, just like it was for her, rather than just wasting her life away in isolation and giving into her own flaws.
